My thoughts on why people want to know:
1) Curiousity - Most of the people curious about whether that girl avatar is actually a girl are just that, curious. They have a gut feeling and they want to see if they're right or not, and thus try to get them on Vent or TS to confirm or deny it - once they know, most don't care anymore and don't act differently; just from my gaming experiences I'd wager about 80% or more are like this.
2) Cyb0r Crowd - This is the next step that some people'd take, going further with the knowledge that a person's actually a girl and seeing if they can develop a relationship with that person. Why? *shrugs* maybe they're ugly in real life and no one likes them, or they prefer gamer girls, or a variety of other reasons, but they want to create and develop a relationship with anyone female. Annoying? Yes, but in EVE you could at least pod them back to the stone age or block them.
3) RL Stalker - the first two are harmless and annoying respectively, but I see this very small minority as actually dangerous. A few girls I've worked with ingame have actually told their friends,corpmates, and myself on forums what's happened to them, from creepy emails to strangers somehow getting an MSN nick of theirs and confessing their love to them, etc. These guys need to get their heads checked and can be very dangerous, and are one of the reasons im my opinion why we don't see more girls in online games. These people want to get some from their ingame "lovers" at any cost .
I fall into the neutral or curiousity category personally. I either don't care, or am curious to see if a hunch I have is right, and then let it be and continue about my business - do us a favor though, if you spot some creep, name and shame him on the forums, I don't want to ever recruit someone like that into the corps or alliances I play with.

Lots of people SAY they don't care, but then spout the "everyone is a man til proven otherwise" nonsense. That means they not only care, they feel they have to have some sort of emotional protection from female avatars.
That's just very weird to me.
Sense when does one gender make sense to the other?

To be brutally honest, some days I wonder if a minority of people actually believe that.
